Investigation of the osmotic response of turbot spermatozoa based on the kinetics of light transmission of incubated cells recorded on a spectrophotometer. more

There are 5 xlsx files with a number of tables 1. File "1_Different_solutions_osmolarities_preliminary" It contains 4 sheets: "male 1", "male 2", "male 3" and "Density, motility, time". The first three sheets ("male 1", "male 2", "male 3") have a similar structure. Measurements of the dependence of light transmission on time were carried out at a temperature of +15°C. This is indicated on each sheet. Each sheet contains 6 tables. Each table consists of two columns and represents the dependence of the light transmission ["T,%"] of the cuvette in which the sperm suspension is incubated versus time ["Time [sec]"] after adding the spermatozoa and brief shaking for homogenization. For the spermatozoa of each male, the kinetics of light transmission was recorded in six media of different osmotic concentrations (sodium chloride): 2000, 1200, 1094, 850, 300 and 0 mOsmol/kg. Sheet "Density, motility, time" contains information on the characteristics of the ejaculates obtained from turbot males: concentration, motility and time of sperm motility, as well as the volume of ejaculate. "preliminary" - means that some part of the data is invalid, because it was obtained during the adaptation of this spectrophotometric method. Data are rounded to 4 decimal places. Since in the future they are used to approximate the functional dependence, the rounding accuracy in this case is of secondary importance. 2.File "2_Different_solutions_osmolarities_temperature" The file contains 9 sheets. They are called "850_15", "2000_22", etc. The first number in the name means the osmolarity of the medium - 850 mOsmol/kg, the second number - the temperature at which the measurements were performed (8, 15 or 22°C). Each sheet contains 9 tables (measurement repetitions). The data is presented similarly to file 1 - this is the kinetics of light transmission measured on a spectrophotometer. 3.File "3_Cryoprotectants" The file contains 5 sheets. They are called "METH", "DMSO", "GLY", "EG", "PG" and mean cryoprotectants methanol, dimethyl sulfoxide, glycerol, ethylene glycol and propylene glycol. Eeach sheet contains 9 tables of the kinetics of light transmission: 3 different temperature measurements in three repetitions. The data is presented similarly to file 1 - this is the kinetics of light transmission measured on a spectrophotometer. Inubation of turbot spermatozoa in one-molar solutions of cryoprotectants prepared in iso-osmotic saline solution. 4. File "4_Cryoprotectants_data" The file contains 4 sheets. The first three sheets (for cryoprotectants "EG", "Meth", "DMSO") have a similar structure. Each sheet contains 9 tables - measurements for 3 ejaculates (3 males) in 3 repetitions at the same temperature. The data is presented similarly to file 1 - this is the kinetics of light transmission measured on a spectrophotometer. Sheet "data" contains clear information about samples before and after cryopreservation (mobility parameters), as well as information about sample codes 5. File "5_Cryopreservation" The file contains 2 sheets. The first leaf is an exposure of cryopreserved sperm in a medium with an osmolarity of 1100 mOsmol/kg: 3 ejaculates (3 males), 5 repetitions each. he data is presented similarly to file 1 - this is the kinetics of light transmission measured on a spectrophotometer. The second sheet is information about cryopreservation (cryo-protocol and spermatozoa motility before and after cryopreservation).
